Miaoling Shared Plasmids (1) | Let-7 miRNA Sponge Lentiviral Vectors Shared by Sun Yat-sen University, China

3 min read

A research team at Sun Yat-sen University, China, has contributed two laboratory-developed and experimentally validated lentiviral vectors targeting the Let-7 family of microRNAs (miRNAs) to the Miaoling Plasmid Sharing Platform. Through this plasmid-sharing initiative, valuable research tools developed in individual laboratories are made available to researchers in China and around the world, helping reduce the time and cost associated with rebuilding existing vectors and supporting further research in related fields.

Both sponge vectors have been described in Signal Transduction and Targeted Therapy, a Nature Portfolio journal (DOI: 10.1038/s41392-021-00497-4). In the study, the research team used the Let-7-5p-Sponge and Let-7-3p-Sponge vectors to antagonize the functions of endogenous mature Let-7 miRNAs in THP-1 cells. The findings demonstrated that Let-7 miRNAs broadly regulate cellular inflammatory responses and participate in the regulation of inflammatory signaling pathways triggered by viral infection.

The article's Data Availability section explicitly identifies the Miaoling Plasmid Sharing Platform as the repository for these two plasmids, making them available to other researchers.

We sincerely thank Sun Yat-sen University for its trust in the Miaoling Plasmid Sharing Platform and for making these independently developed research plasmids available to the broader scientific community. Both plasmids are now officially listed on the Miaoling platform and available for researchers to order.

Plasmid Descriptions

pLVX-AcGFP-N1-Let-7-5p-Sponge (P20227)

Built on the pLVX-AcGFP-N1 lentiviral backbone, this vector carries a Let-7-5p miRNA sponge sequence, along with an AcGFP green fluorescent reporter and an ampicillin resistance marker.

The sponge sequence competitively binds endogenous mature Let-7-5p miRNA within cells, reducing its ability to repress downstream target genes.

The vector can be introduced directly by transient plasmid transfection or packaged into lentiviral particles for delivery into hard-to-transfect cell types, including primary cells and stem cells. AcGFP fluorescence enables visual monitoring of transduction or reporter expression. When combined with an appropriate selection strategy, the vector can support the generation of stable cell models with sustained inhibition of Let-7-5p function.

Recommended competent cells: Stbl3. Culture at 37°C.

This vector may be useful for mechanistic studies investigating the roles of Let-7-5p in cell proliferation, cancer biology, and development.

pLVX-AcGFP-N1-Let-7-3p-Sponge (P20228)

This vector also uses the pLVX-AcGFP-N1 lentiviral backbone, but carries a Let-7-3p miRNA sponge sequence, together with an AcGFP fluorescent reporter and an ampicillin resistance marker.

The sponge sequence is designed to competitively bind endogenous mature Let-7-3p miRNA, thereby reducing Let-7-3p-mediated gene silencing.

The vector can be used for transient transfection or packaged into lentiviral particles to establish stable cell models with sustained inhibition of Let-7-3p function. AcGFP fluorescence provides a convenient means of tracking reporter-positive cells.

Recommended competent cells: Stbl3. Culture at 37°C.

This vector may be useful for investigating the biological functions of Let-7-3p in cell differentiation and disease models.

Research Tip: Why Study Let-7-5p and Let-7-3p Separately?

Let-7-5p and Let-7-3p are mature miRNA products derived from opposite arms of the same miRNA precursor. Although they originate from the same precursor, they can have distinct target profiles and biological functions.

Using the two sponge vectors separately or in parallel can help researchers distinguish the contributions of each mature strand to specific biological phenotypes. This distinction was also important to the experimental approach described in the Signal Transduction and Targeted Therapy study.

Miaoling's Research Plasmid-Sharing Program

The Miaoling Plasmid Sharing Platform welcomes contributions from research groups at universities and scientific institutes that have constructed and validated their own plasmid vectors.

After submitted plasmids pass quality control, they can be added to Miaoling's in-stock inventory, making these research tools accessible to a broader scientific community. The initiative aims to promote the open sharing of molecular biology resources, reduce duplicated effort, and improve research efficiency.

If your laboratory is interested in sharing plasmids, please contact the Miaoling team to submit the relevant vector information and bacterial strains. We will assist with the subsequent quality-control and product-listing process, helping make valuable research tools available to fellow scientists.
